Cleanout blocks go in the bottom course of any wall you intend to fill with concrete. As the wall goes up, mortar drops down the cores and sits on the footing - if it isn't removed the corefill won't bond properly, and your engineer or certifier will want to see the starter bars before you pour anyway. The removable panel at the base lets you clean out the debris and lets the inspector look in. Once you're signed off you fit the matching spring-and-biscuit closer (code 50.45), reinstate the panel and fill the cores. You need one at every core that's being filled.
